Anil-Babesia rodhaini Monoclonal Antibodies: Effect against Babesia rodhaini and Cross-reactivity with Human Starin of Babesia microti OAK

Anti-Babesia rodhaini monoclonal antibodies (mAb), namely: 1-E7, 2-H2 and 3-B8, significantly suppressed the development of high parasitemia in BALB/c mice infected with B. rodhaini and all mAb-treated mice survived the infection. While, only monoclonal antibody 3-B8 showed some inhibitory effect against Babesia microti, with mice showing high parasitemia of 18.04 ± 2.69 %, at 9 days post-exposure. Westernblot analysis of B. rodhaini and B. microti parasite extract reacted with anti-B. rodhaini monoclonal antibodies showed cross-reactive bands of molecular weights 62 and 55 kilodaltons. Cellualr Subsets Involved in Protective Immunity to Babesiarodhaini Infection in BALB/c Mice OAK

Protective immune responses and the functional role of spleen cells in mice infected with Babesia rodhaini were examined with an in vitro proliferation assay systems and by in vivo passive transfer of spleen cells to uninfected mice. Mice that resolved primary babesial infection after chemotherapy (Babesia immune mice) had transient and low parasitemia after challenge infection and high rates (75%) of survival. Babesia hyperimmune mice, by contrast, had no detectable parasitemia after challenge and 100% survival. Neospora caninum infection is an important disease affecting bovine and canine populations worldwide. The dense granule antigen 7 of N. caninum (NcGRA7) is considered to be an immunodominant antigen. In the present study, the gene encoding truncated NcGRA7 (NcGRA7t) lacking an N-terminal signal peptide was expressed in Escherichia coli, and its diagnostic potential in an enzyme-linked immunosorbent assay (ELISA) was evaluated. The ELISA could clearly discriminate between known N. caninum-positive and -negative sera from dogs. High Genetic Diversity of Anaplasma marginale Detected from Philippine Cattle OAK

A total of 658 cattle in 6 provinces in the Philippines were screened for Anaplasma marginale infection by using a diagnostic heat-shock operon (groEL) gene-PCR assay. The screening-positive samples were further tested using the major surface antigen protein 1a (Msp1a) gene-PCR assay. Screening PCR results showed 130 cattle (19.8%) were positive for the A. marginale infection. Subsequent amplification using the Msp1a gene only showed 93 samples (14.1%) to be positive. In addition, 37 tandem-repeat structures, including 20 novel structures, and 41 distinct genotypes were identified. Surveillance of the presence of larval mosquitoes was performed in the suburban municipality of Sta. Ignacia, Tarlac Province, Philippines, with approximately 94.0% of the land area still devoted to agriculture. Five districts were covered, namely: Macageuing, San Francisco, San Vicente, Sta. Ines Centro, and Sta. Ines East. The standard dipping method was used to collect mosquito larvae from rice fields, ponds, creeks, streams and used tires and larvae were identified at the genus level. Serological survey of Toxoplasma gondii infection was carried out in 68 urban (Manila) and 72 suburban (Dasmariñas, Cavite) residents, comprising 69 females and 71 males, age 16-56 years old. Using a survey questionnaire, the name and address, gender, age, weight, height, civil status, cat exposure, and additional information like history of pregnancy and current reproductive status of females were obtained. The presence of anti-T.gondii antibodies was confirmed using the Toxocell latex agglutination test.
